Separation and Purification of Baicalin Using Deep Eutectic Solvent and Surfactant

Abstract: The deep eutectic solvents (DESs) are used to replace the traditional solvents and a new method is established for the separation of baicalin by the eutectic solvent-cloud point separation (DESs-CPE) system. Many variables including the type of DESs in the extractants, the water content in the DES, the solid-liquid ratio, the extraction temperature and time are evaluated and optimized. Two steps of cloud point separations are used to separate and recover baicalin to remove DESs. Quantitative and qualitative analysis of baicalin is carried out by UPLC. Under optimized experimental conditions, the maximum extraction efficiency of baicalin is (124.73±3.81) mg/g, about 1.64 times of the traditional method. After two cloud point separations, the maximum recovery rate of baicalin is (72.46±0.72)% and the purity is (89.72±1.14)%.
