Electrospray Ionization Tandem Mass Spectrometry on the Interaction of Baicalin and Glutathione

Abstract: The interaction between baicalin and glutathione was studied by electrospray ionization multiple-stage tandem mass spectrometry. The results show that the oxidation product of baicalin can interact with glutathione. Adduct of glutathione and baicalin is unstable and further undergoes reactions such as auto-oxidation, hydration or deuronic acid. The binding site of baicalin on glutathione is identified at the C8 position of baicalin aglycone. These results provide insights of the interaction of glutathione with baicalin at molecular level without the need of the complicated sample separation and purification processes. This strategy can be applied to investigate the interaction of other natural products and peptides.
