一种近红外快速响应型硫化氢荧光探针的合成及细胞成像

Synthesis and Cell Imaging of a Near-Infrared Fast Responsive Hydrogen Sulfide Fluorescent Probe

摘要:

基于CuS的生成,设计合成了一个含氟硼二吡咯(Boron-dipyrromethene, BODIPY)的硫化氢荧光探针Mito-BPN-Cu。 光物理性质研究发现,探针的最大吸收峰为694 nm,最大发射波长为805 nm。 Mito-BPN-Cu对硫化氢表现出高的选择性,且具有极快的响应速度(10 s内)。 在纯乙腈体系及V(乙腈)∶V(水)=1∶1体系中,Mito-BPN-Cu对硫化氢(H2S)均具有较高的灵敏性,其中在后者溶剂体系中的检测限达到了0.56 μmol/L。 Mito-BPN-Cu具有低的细胞毒性,在48 h、最大浓度150 μmol/L下,HCT116细胞的存活率仍大于80%。 此外,该探针能特异性靶向线粒体,皮尔森相关系数(Pearson′s correlation coefficient, PCC)为0.92。 鉴于该探针上述优点,Mito-BPN-Cu成功用于对HCT116细胞中的硫化氢荧光成像。

Abstract:

Based on the formation of CuS, a fluorescent probe of H2S containing BODIPY, Mito-BPN-Cu, was designed and synthesized. The study of photophysical properties show that the maximum absorption peak and the maximum emission wavelength are 694 nm and 805 nm respectively. Mito-BPN-Cu shows high selectivity for hydrogen sulfide and has an extremely fast response speed (within 10 seconds). In both pure acetonitrile and V(acetonitrile)∶V(water)=1∶1 solvent systems, Mito-BPN-Cu has high sensitivity to H2S, and the detection limit in the latter solvent system reaches 0.56 μmol/L. Mito-BPN-Cu showed low cytotoxicity, and the survival rate of HCT116 cells was still greater than 80% at 48 h and even the concentration of the probe was 150 μmol/L. In addition, the probe can specifically target mitochondria with a Pearson correlation coefficient of 0.92. In view of the above advantages of the probe, Mito-BPN-Cu was successfully used for fluorescence imaging of hydrogen sulfide in HCT116 cells.
