Effect of Electric Field-Regulating Cholesterol-based Liquid Crystal Films on the Growth and Differentiation of Fibroblasts

Abstract:

The liquid crystal ordering of cholesterol-g-polylactic acid oligomer (CLA) film is regulated by external direct-current electric field, and its effects on the adhesion, proliferation and differentiation of fibroblasts are evaluated. The liquid crystalline properties of CLA polymer aredetected by differential scanning calorimeter. The CLA films is prepared under a direct-current electric field with different intensities, and the liquid crystal ordering is detected by X-ray diffractometer and polarized light microscope. The adhesion and proliferation of fibroblasts on the CLA films are detected by a Cell Counting Kit-8 kit. The expression of α-smooth muscle actin (α-SMA) is detected by immunofluorescence staining and western blotting. The results show that CLA polymer has liquid crystal property, and the external electric field could regulate the liquid crystal property of CLA film through inducing the arrangement of CLA molecules. The enhancement of liquid crystal property of CLA films isbeneficial to the proliferation of fibroblasts and the expression of α?-SMA protein. These results suggest that electric field could promote the phenotypic transformation of fibroblasts to myofibroblasts by increasing the liquid crystal ordering of CLA polymer film, without changing the composition of matrix materials.
