电喷雾串联质谱研究黄芩苷与谷胱甘肽的相互作用

摘要: 建立了研究黄芩苷和谷胱甘肽的相互作用的电喷雾多级串联质谱方法,结果表明黄芩苷首先自氧化生成黄芩苷半醌,随后黄芩苷半醌与谷胱甘肽进一步发生反应,生成谷胱甘肽与黄芩苷加合物,该化合物不稳定,可进一步发生自氧化、水合反应或脱糖醛酸等反应。 利用串联质谱技术鉴定了谷胱甘肽与黄芩苷的结合位点是在黄芩苷苷元的C8位。 该方法从分子水平揭示了黄芩苷和谷胱甘肽的相互作用过程,无需复杂的样品分离提纯过程。 研究结果不仅有助于深入理解黄芩苷在癌细胞内的代谢研究,而且还可以应用于其他天然有机小分子和小肽相互作用的研究。

Abstract: The interaction between baicalin and glutathione was studied by electrospray ionization multiple-stage tandem mass spectrometry. The results show that the oxidation product of baicalin can interact with glutathione. Adduct of glutathione and baicalin is unstable and further undergoes reactions such as auto-oxidation, hydration or deuronic acid. The binding site of baicalin on glutathione is identified at the C8 position of baicalin aglycone. These results provide insights of the interaction of glutathione with baicalin at molecular level without the need of the complicated sample separation and purification processes. This strategy can be applied to investigate the interaction of other natural products and peptides.
