甘草酸二铵与牛血清白蛋白相互作用的光谱

Spectroscopic Study on the Interaction between Diammonium Glycyrrhizinate and Bovine Serum Albumin

摘要:

在模拟人体生理条件下,用常规荧光光谱和紫外-可见吸收光谱研究甘草酸二铵和牛血清白蛋白的相互作用机制,并利用同步荧光光谱和三维荧光光谱研究了药物对牛血清白蛋白构象的影响。研究表明,甘草酸二铵对牛血清白蛋白具有荧光猝灭作用,且为单一的动态猝灭过程。根据Stern-Volmer方程求出了两者相互作用的动态猝灭常数,并根据Förster非辐射能量转移理论确定了药物与蛋白的结合距离。

Abstract:

The interaction between diammonium glycyrrhizinate and bovine serum albumin was studied in physicological solution by spectroscopic methods including fluorescence spectra and UV–visible absorption spectra. Synchronous fluorescence and three-dimensional fluorescence spectra were used to investigate the conformation change of bovine serum albumin due to the addition of diammonium glycyrrhizinate. It showed that the dynamic quenching exited between diammonium glycyrrhizinate and bovine serum albumin. Based on the equation of Stern-Volmer, the quenching constant was obtained. According to Förster theory of non-radiation energy transfer, the distance between bovine serum albumin and diammonium glycyrrhizinate was determined.
