The methylated oligosaccharides products were characterized by ultra performance liquid chromatography triple quadrupole tandem mass spectrometry (UPLC-QQQ-MS). The solid phase methylation conditions were further optimized based on previous work. Six reducing oligosaccharides (maltose, maltotriose, maltotetraose, maltopentaose, maltohexaose and maltoheptaose) and three non-reducing oligosaccharides (sucrose, kestose and nystose) were detected in ginseng. This method could be applied in the quantitation of nine oligosaccharides in ginseng, simultaneously. UPLC-QQQ-MS combined with solid phase methylation could be used to efficiently derive and quantify oligosaccharides in ginseng. Therefore, an effective method is devdeloped to investigate the influence of growth environment on the distribution of oligosaccharides in ginseng. %U http://yyhx.ciac.jl.cn/EN/10.19894/j.issn.1000-0518.200240
