HIV感染者头发中糖皮质激素和内源性大麻素的高效液相-串联质谱检测

Detection of Endocannabinoids and Glucocorticoids in Hair of People Living with HIV by High Performance Liquid Chromatography Tandem Mass Spectrometry

摘要: 建立了同时检测头发中皮质醇、可的松、脱氢表雄酮、花生四烯酸乙醇氨、1-花生四烯酸甘油、油酰乙醇胺、十六酰胺乙醇和硬脂酸单乙醇酰胺这8种物质的方法,并用于检测58名艾滋病病毒(HIV)感染者头发样本中的目标物含量,探讨HIV感染对这些物质含量的影响。 使用甲醇对头发中待测物进行提取,固相萃取柱对样品提纯;采用大气压化学电离离子化源,在正离子和多反应监测模式下进行定量检测。 在优化的色谱条件下,8种待测物分离良好,峰形对称。 8种分析物的标准曲线相关系数均大于0.99;定量限在1.0~25 pg/mg;回收率在(100±15)%范围内;日内日间变异系数小于15%;归一化基质因子接近1;稳定性、残留效应均符合检测标准要求。 HIV感染人群头发中皮质醇含量比正常健康人群更高(P<0.01),但花生四烯酸乙醇氨含量更低P<0.001),表明感染者的内源性大麻素系统可能出现异常,下丘脑-垂体-肾上腺轴的负反馈调节能力不足。

Abstract: A method for simultaneous detection of cortisol, cortisone, dehydroepiandrosterone, arachidonoyl ethanolamide, 1-arachidonoyl glycerol, palmitoyl ethanolamide, oleoyl ethanolamide, stearoyl ethanolamide based on high performance liquid chromatography-tandem mass spectrometry was developed. Furthermore, methanol was used to extract target analytes in the hair and solid phase extraction was used to purify the sample. The simultaneous quantitation was conducted by using atmospheric pressure chemical ionization source with the positive mode and multi-reaction monitoring mode. Under optimized chromatographic conditions, the peak shapes of the 8 analytes are sharp, symmetrical and well-separated. The results indicate that the correlation coefficients of standard curves are greater than 0.99 and limits of quantitation for the eight analytes are among 1.0~25 pg/mg. The recovery is within (100±15)% and intra- and inter-day coefficient variations are less than 15%. The normalized matrix factors are close to 1. The stability and carry over also meet the criterion. The method has good stability and reproducibility. The population analysis results show that the content of cortisol in the hair of HIV-infected people is higher than that of healthy people (P<0.01), and the content of arachidonoyl ethanolamide is lower (P<0.001), which suggests that the patient's endocannabinoid system may be abnormal and the hypothalamic-pituitary-adrenal axis has insufficient negative feedback regulation ability.
